Glass slide:
1. Definition: Slides containing biological specimens or slices.
2. Material: Glass slides are used to place experimental materials in the experiment. They are rectangular, thick and have good light transmittance. The smear materials include single-cell organisms, small algae, blood, bacterial culture medium, loose tissues of animals and plants, testis, anthers and so on.
When smearing, attention should be paid to: (1) The slide must be cleaned. (2) The slide should be flat. (3) The coating should be uniform. Apply the droplets to the right in the middle of the slide. Apply them evenly with the edge of the dissecting knife or toothpick. (4) The coating should be thin. Using another slide as pusher, the slide with smear solution dropping along the surface (the angle between the second slide should be 30 - 45) is pushed gently from right to left and coated into a uniform thin layer. (5) Fixation. If fixed, chemical fixatives or drying (bacteria) can be used. (6) Staining. Bacteria use methylene blue, blood use Wright's stain, and sometimes iodine can be used. The dye solution should cover all the coatings. (7) Rinse. Dry or bake with absorbent paper. (8) Seals. Canadian gum seals for long-term preservation.
<2> Compression is a method of making bio-materials by placing them between slide and cover and applying certain pressure to compress and disperse tissue and cells.
<3> Filling method is a method of making slide specimens by whole sealing of biomaterials. Temporary or permanent filling can be made by this method.
Film materials include: micro-organisms such as chlamydia, sponge, amoeba, nematode; hydra, plant leaf epidermis; insect wings, feet, mouthparts, human oral epithelial cells, etc.
Attention should be paid to the fabrication of mounting method: (1) When carrying glass in hand, attention should be paid to keeping it flat or putting it on the platform. When dripping, the amount of water should be appropriate to be covered by the cover glass. (2) The material should be unfolded with an anatomical needle or tweezers so as not to overlap and flatten on the same plane. (3) When covering the glass slide, cover the water droplets slowly from one side to prevent bubbles from appearing. (4) When dyeing, a drop of dyeing solution is placed on one side of the cover glass and absorbent paper is used to attract the specimen from the other side, so that the specimen under the cover glass can be uniformly colored. After dyeing, drop a drop of clear water in the same way, and then suck out the dye solution and observe it under a microscope.
Slices are slide specimens made of thin slices cut from organisms.
Due to different requirements, the blade can be used for bare-handed slicing, tissue blocks can also be embedded in paraffin or collodion or frozen at low temperature, and sliced with a slicer. Cut into 5-10 micron slices for optical microscopy. Ultrathin sections made of epoxy resin or methacrylic acid embedded tissue blocks with a thickness of 20-50 nanometers are specially observed under electron microscopy. Paraffin sections are commonly used in teaching, such as root tips and stems.
5. Cleaning method: Wash it with water or alcohol, then wipe it with cotton or gauze (it is better to wipe the mirror paper and avoid touching the slide with your finger, so as not to leave fingerprints on it, which will affect the next observation and use).
